  80. 1) My laboratory is engaged in studies of viral latency using murine cytomegalovirus, a member of the family of beta-Herpes viruses. Of particular interest in our laboratory, in 1993, has been the application of PCR to detect viral messenger RNA in latently infected animal cells. This questions is, we believe, a biologically very important one since it reflects directly on the issue of the state of activity of the viral genome in latent infection.
  81. 2) In these experiments using reverse transcriptase PCR we were able to detect immediate-early I transcripts at a level roughly 10-fold higher than that of viral genome, indicating that a significant fraction of viral genomes is transcriptionally active in latency. These findings may be indicative of an abortive infection in which transcription is limited to the immediate-early genes, or the result of a very low level persistent infection, or a transient reactivation of latent virus. To further assess the state of viral activity in latent infection we have prepared four hybridomas from the splenocytes of latently infected animals using standard techniques. We then tested the activity of the resultant monoclonal antibodies against proteins expressed from purified clones derived from a cDNA which we developed. We have further established the gene map location encoding a unique protein and is distinct from the immediate early genes. We have sequenced this gene and performed in situ hybridization on acutely latently infected tissues. Further studies using immunohistochemistry indicate that this viral gene is active during latency although not exclusively. We are currently in the process of attempting to further characterize this protein and its importance, if any, in the control of viral latency.
  82. 3) In addition, I have been very active in the conduct of clinical trials relevant to HIV/AIDS. These include a study of early versus late AZT of which I was co-principal investigator in this VA Cooperative Study and I am principal investigator for a second VA Cooperative Study of HIV Vaccine in Intravenous Drug Users and Their Sexual Partners.
