The transfectability of the hepatocyte spheroids in comparison with hepatocyte monolayer was investigated using a novel non-viral polycationic gene carrier, polyphosphoramidate containing diethylamine side chains (PPA-DEA). The optimal window for transfection after cell seeding and culture was also determined. Using ELISA and HPLC techniques, hepatocyte-specific functions such as albumin secretion and P450 activity were measured. PPA-DEA was found to be a viable non-viral gene carrier since it produces reasonable transgene expression and has no adverse effects on the hapatocyte-specific functions.Keywords:
