Abstract: A novel method for sensitive environmental control of the molecular recognition process between a protein receptor and its ligand is presented. The method is based on the conjugation of a smart, temperature-sensitive polymer, poly(N-isopropylacrylamide), to a specific genetically-engineered site, thiol. It was shown that the site-specific conjugation of a temperature-sensitive polymer to a genetically engineered protein provides a very sensitive temperature control of the ligand-protein recognition process. Such molecular switches could have widespread applications in medicine and biotechnology.
